Activation of Smad signaling enhances collagenase-3 (MMP-13) expression and invasion of head and neck squamous carcinoma cells

Abstract: Squamous cell carcinoma (SCC) cells of the head and neck specifically express collagenase-3 (matrix metalloproteinase-13 (MMP-13)), the expression of which correlates with their invasion capacity. Transforming

“…32 SDF-1 binding to CXCR4 can activate Akt, 34 transactivate epidermal growth factor receptor, 35 activate JAK2/STAT3 leading to enhanced survival and inhibition of apoptosis 36 and alter ras signaling. 37 SDF-1 signaling also induces production of the matrix metalloproteinases MMP-2, MMP-9 38 and MMP-13 39 that are important for breaking down basement membrane.…”

“…The MMPs can be divided into subgroups of collagenases, gelatinases, stromelysins, matrilysins, membrane-type MMPs, and other MMPs according to their substrate specificity and function (Yong et al 2001). MMP-13 is a collagenolytic MMP characterized by a wide substrate specificity, and it is expressed by many types of invasive tumors, such as breast carcinomas (Freije et al 1994), squamous cell carcinomas of the head and neck (Leivonen et al 2006). The correlation between MMP-13 expression level and invasive capacity suggests that MMP-13 is an important molecule for cancer progression.…”

“…Increased expression of VEGF is seen in OSCC through all stages of disease progression [106][107][108]. Other factors that promote blood vessel formation and endothelial cell migration include, prostaglandin E2 (PGE 2 ) and TGF-β̣ are often overexpressed by tumor cells [109][110][111][112][113]. COX-2 inhibition has been shown to be effective in controlling OSCC cell proliferation and this is shown to be mediated specifically through the inhibition of PGE 2 .…”
