Activation of Monocyte and Granulocyte Antibody-Dependent Cytotoxicity by Phorbol Myristate Acetate

Abstract: We have characterized the effects of phorbol myristate acetate (PMA) on human monocyte and neutrophil oxidative metabolism and antibody-dependent cytotoxicity toward anti-D sensitized human erythrocytes (RBC) and a human lymphoblastoid cell line (CEM). Hexose monophosphate shunt activity was measured by [1- 14 C]glucose oxidation and target lysis by 51 Cr release. PMA produced a dose-dependent stimulation of hexose monophosphate shunt activity. Neutrophils respon… Show more

“…Many of the reports listed in Tables I and II discuss the anticancer effect of PMN via ADCC,27, 41, 46, 50, 64, 83, 116, 120, 122–126, 128, 129, 131–177 and many provide direct evidence of this antibody‐mediated antitumoral role of PMN in vitro. The first reports were published in 1974 and 1975 by Gale and Zighelboim 131, 132.…”

“…Interferon and PMA were previously reported to enhance PMN-mediated ADCC (14,18), and PMA was reported to stimulate PMN-mediated killing of tumor cells, erythrocytes, and endothelial cells (25,40). We found that the addition of interferon a in concentrations ranging from 5 to 500 IU/ml failed to enhance PMN-mediated killing of unsensitized and antibody-sensitized, VZV-infected targets (Ihara et al, unpublished data).…”
