Activation of large conductance potassium channels inhibits the afferent and efferent function of airway sensory nerves in the guinea pig.

Abstract: Sensory nerves play an important role in airway disease by mediating central reflexes such as cough, and local axon reflexes resulting in the peripheral release of neuropeptides. We have tested whether the benzimidazolone compound, NS1619, an opener of large conductance calcium-activated potassium (BK Ca ) channels, inhibits the activity of sensory fibers, and central and local airway reflexes in guinea pig airways. In in vitro single fiber recording experiments, NS1619 applied to identified receptive fields i… Show more

“…The measurement of cough in conscious guinea pigs has been described previously [14][15][16][17]. Terbutaline (0.05-3.0 mg?kg body weight -1 and 1 mL?kg body weight -1 intraperitoneally) or vehicle (0.5% methylcellulose/0.2% Tween 80 in saline) was administered 30 min prior to exposure to capsaicin (1610 -4 M; 5 min) or citric acid (0.3M; 10 min), and the number of coughs over a 10-min period was recorded.…”

“…They play a key role in the control of neuronal excitability, hormone secretion, and smooth muscle tone (Robitaille et al, 1993;Nelson et al, 1995;Lingle et al, 1996;Fox et al, 1997). Thus, BK Ca channels are considered to be molecular integrators of biochemical and electrical signals.…”

“…They play a key role in the control of neuronal firing (3,4), cell secretion (5), and smooth muscle tone (6). Maxi-K channel activity is under a complex metabolic control that may involve G-proteins (7,8), intracellular Ca 2ϩ concentration (6), and a balance between phosphorylation/dephosphorylation mechanisms (9,10).…”
