Activation of human monocytes by Streptococcus mutans serotype f polysaccharide: immunoglobulin G Fc receptor expression and tumor necrosis factor and interleukin-1 production

Abstract: Streptococcus mutans serotype f polysaccharide (poly f) was prepared from S. mutans whole cells by autoclaving. The poly f was purified by chromatography on DEAE Trisacryl M and Bio-Gel P100, treated with insoluble pronase, and resubjected to chromatography on DEAE Trisacryl M. Normal human blood monocytes, stimulated in vitro with purified poly f, produced extracellular tumor necrosis factor alpha (TNF-alpha) and interleukin-1 beta (IL-1 beta) in a dose-dependent fashion as determined by a heterologous two-si… Show more

“…2). These results are similar to other reports showing that S. mutans stimulates the production of TNF-a and IL-1b in human peripheral blood mononuclear cells or human monocytes, or the S. mutans OMZ175 protein I/II induces the production of TNF-a and IL-1b in human synoviocytes (Benabdelmoumene et al, 1991;Soell et al, 1994;Jiang et al, 1999;Gourieux et al, 2001). Nuclear factor-jB is a multiunit transcription factor that plays a key role in the induction of genes for proinflammatory cytokines (Collart et al, 1990;Ziegler-Heitbrock et al, 1993;Neff et al, 2001) as well as many other immunoregulatory genes (Baeuerle, 1991).…”

“…Antisera production. Polyclonal anti-SR, anti-rhTNF-cx, and anti-rhIL-1 3 sera were raised in rabbits by several injections of 50 tLg of either SR or TNF-ot or IL-1B suspended in Freund's incomplete adjuvant (5). The IgG fractions of anti-SR, anti-rhTNF-ot, and anti-rhIL-13 sera were purified by chromatography on DEAE Trisacryl M essentially by the method of Corthier et al (8).…”

“…It has been postulated that acute inflammation and subsequent tissue damage could result from the deposition of either streptococcal antigens or antigen-antibody complexes within the tissues (37) or from the stimulation of the host immune response by certain antigens, leading to the synthesis of either antibodies that cross-react with host tissues (20) or cytokines, which are believed to play an important role in these chronic inflammatory diseases. We recently showed that serotype polysaccharide from Streptococcus mutans OMZ 175 (serotype f), a member of the viridans streptococci, cah stimulate human monocytes to release tumor necrosis factor alpha (TNF-ot) and interleukin 1 (IL-1,B) (5).…”

“…Although the role of cytokines in the pathogenesis of dental caries is not distinct, proinflammatory cytokine production is induced by components of S. mutans. 7,8 The aim of this study was to assess association between IL-1α and IL-6 levels and dental caries, especially in S. mutans infections. In this study, 70 patients with dental caries were selected, who referred to the Department of Oral and Maxillary Surgery, Faculty of Dentistry, Tabriz University of Medical Sciences.…”
