Activation of Epithelial-Mesenchymal Transition and Altered β-Catenin Signaling in a Novel Indian Colorectal Carcinoma Cell Line

Abstract: Colorectal cancer is the third major cause of cancer-related mortality worldwide. The upward trend in incidence and mortality rates, poor sensitivity to conventional therapies and a dearth of early diagnostic parameters pose a huge challenge in the management of colorectal cancer in India. Due to the high level of genetic diversity present in the Indian population, unraveling the genetic contributions toward pathogenesis is key for understanding the etiology of colorectal cancer and in reversing this trend. We… Show more

“…Higher expression level of β-catenin was also achieved over time when HCEC-1CT were exposed to exosomes derived from metastatic SW620, and its localization into the cell nuclei was observed after three-day incubation. Aberrant Wnt signaling is a prominent feature of several cancers, especially CRCs, which was found in 90% of cases to exhibit overexpression of β-catenin [ 38 , 39 ].…”

“…Western blotting analysis was performed for total proteins extracted from exosomes derived from CaCo-2 and SW620 and for HCEC-1CT cells, untreated or treated with exosomes derived from both cancer cell lines, as previously described [ 39 ]. Untreated cells maintained in culture medium for 24 h were used as control.…”

“…β-catenin is sequestered and maintained by cadherins at adherens junctional complexes [56]. However, once EMT or EndMT is activated, β-catenin takes on a new role as a signaling molecule for the Wnt pathway as it disassociates from the cadherins at the membrane, translocates to the nucleus and binds to the TCF/LEF family of transcription factors to activate EMT and EndMT transcription factors including Snai1 [57,58]. Computational modeling of transcription factors regulating EMT in RPE identified lymphoid enhancer-binding factor 1 (LEF-1) as one of the candidate nodes in the EMT transcriptional regulatory network of RPE [59].…”

“…EMT is an important mechanism that affects the invasion and metastasis of CRC ( 21 ). Several growth factors, such as TGF-β and epidermal growth factor, mediate EMT ( 22 , 23 ). Notably, the results of the present study demonstrated that TGF-β induced miR-325-3p expression.…”
