Activation of CREBZF Increases Cell Apoptosis in Mouse Ovarian Granulosa Cells by Regulating the ERK1/2 and mTOR Signaling Pathways

Abstract: CREBZF, a multifunction transcriptional regulator, participates in the regulation of numerous cellular functions. The aims of the present study were to detect the localization of CREBZF expression in the ovary and explore the role of CREBZF and related mechanisms in the apoptosis of ovarian granulosa cells. We found by immunohistochemistry that CREBZF was mainly located in granulosa cells and oocytes during the estrous cycle. Western blot analysis showed that SMILE was the main isoform of CREBZF in the ovary. … Show more

“…Apoptosis of ovarian cells, to a certain extent, is closely related to the atresia of follicles [ 39 ], and it is adjusted by a complex regulatory network [ 40 ]. Therefore, studies on the apoptosis and corresponding action mechanism of CCs may be beneficial to elaborate the follicular development and reproductive characteristics of yak.…”

“…The cumulus-oocyte complex and granulosa cells were released using a 1 mL syringe needle by puncturing the dominant follicles (>200 μm in diameter) on the ovaries [ 39 , 40 ]. The FSH receptor was used to identify the purity of the ovarian granulosa cells [ 41 ]. After the oocytes were aspirated, the granulosa cells were filtered through a sieve with a pore size of 0.074 mm and seeded into a 6-well plate.…”

“…SMILE is a multifunctional transcription factor that reduces hyperglycemia induced by CREB/CRTC2 signaling (31) and represses adipogenesis by regulating peroxisome proliferatoractivated receptor g (PPARg) (32). CREBZF, the long form of SMILE, inhibits the ERK1/2 and mTOR signaling pathways by phosphorylating ERK1/2 and p70 S6 kinase (S6K1) and activates autophagy by increasing the LC3-II level in ovarian granulosa cells (33). Moreover, CREBZF stabilizes and activates the tumor suppressor p53 and protects against ultraviolet-induced cell death (34) PBMCs from patients with UC with severe inflammation.…”
