Activation of Coagulation by Lenalidomide-Based Regimens for the Treatment of Multiple Myeloma

Abstract: We investigated the procoagulant effects of lenalidomide (Len)-based regimens in vitro focusing on tissue factor (TF) and phosphatidylserine (PS). We examined the effects of a pharmacological concentration of Len with or without the corticosteroid dexamethasone (Dex) and the proteasome inhibitor bortezomib (Bor) using the human vascular endothelial cell line EAhy926 and the monocytic cell lines THP-1 and U937. Cell-surface procoagulant activity (PCA) was induced by Dex-containing regimens in all lines. Express… Show more

“…It also causes apoptosis, leading to the phosphatyl-serine induced activation of procoagulant tissue factor on cell membranes thus making these cells more thrombogenic. [12] However, it was seen that incidence of VTE in MM doesn't increase solely with Thalidomide. [13] When Thalidomide is combined with chemotherapeutic agents such as Doxorubicin or melphalan with prednisone, there was a sharp increase in the incidence of VTE [14,15].…”

“…We avoided freeze-thaw cycles. After incubation at 37°C for 3 min, 50 μL of 25 mM calcium chloride was added, and the plasma recalcification time was measured using a semi-automatic coagulator (CA-50; Sysmex, Kobe, Japan) [ 12 , 13 ]. CA-50 detects a change in plasma optical density during blood coagulation, that is, fibrin formation.…”
