Activation of Caspase-2 in Apoptosis

Abstract: Members of the CED-3/interleukin-1␤-converting enzyme (ICE) protease (caspase) family are synthesized as proforms, which are proteolytically cleaved and activated during apoptosis. We report here that caspase-2 (ICH-1/NEDD-2), a member of the ICE family, is activated during apoptosis by another ICE member, a caspase-3 (CPP32)-like protease(s). When cells are induced to undergo apoptosis, endogenous caspase-2 is first cleaved into three fragments of 32-33 kDa and 14 kDa, which are then further processed into 18… Show more

“…Although it has been reported that caspase-3 may be involved in the process of caspase-2 activation (Li et al, 1997), our kinetic observations suggest that caspase-2 activation was associated with early activation of caspase-3 in TGFb-activated BL41 cells. However, no inhibition of caspase-3 activation and PARP cleavage were observed in the presence of the caspase-2 speci®c inhibitor ZVDVAD-fmk.…”

“…Our results also suggest that caspase 2 activation occurs after the activation of caspase 8 but before the activation of the mitochondrial pathway. A number of studies have found that caspase 3 can directly activate caspase 2, 44,45 which has led some investigators to conclude that caspase 2 can function downstream of the mitochondria. On the contrary, other studies have implicated caspase 2 in the activation of the mitochondrial pathway, either directly or indirectly by facilitating the cleavage of Bid.…”

“…At the time of sampling this patient was Binet stage A, but in the last year has progressed to stage C. Activation of caspase-2 during the execution phase of apoptosis has been observed in different tumor cell lines. 21,30,31 The precise mechanism by which procaspase-2 is activated in cells is not known. It may involve recruitment through an adapter such as RAIDD/CRADD, 32,33 or based on activities of recombinant enzymes, it has been proposed that caspase-3 activates pro-caspase-2.…”

“…21,30,31 The prodomain of caspase-2 binds to the adaptor molecule RAIDD/CRADD which also binds the receptor-interacting protein RIP and may thus regulate apoptosis. 32,33 It is not known which if any intracellular substrates are cleaved by caspase-2 in apoptosis.…”
