Activation of adenylate cyclase in cdc25 mutants of Saccharomyces cerevisiae

Abstract: The activation of adenylate cyclase by guanine nucleotides and 6-deoxyglucose was studied in membrane preparations from S. cerevisiae mutants lacking the CDC25 gene product. Adenylate cyclase from cdc25 ts membranes was activated by GTP and GppNHp in membranes from cells collected after glucose was exhausted from the medium. The activation was also observed in membranes from repressed cells at 2.5 mM MgZ'. It is also shown that 6-deoxyglucose can activate adenylate cyclase in the absence of CDC25 gene product.… Show more

“…We have previously shown that intracellular acidi¢cation causes an increase in the GTP/GDP ratio on the Ras proteins through inactivation of the Ras-GTPases, Ira1 and Ira2 [17]. In addition, Cdc25 and Ras are also involved in membrane targeting of adenylate cyclase [60,61]. As such, the ampli¢cation model would be consistent with several studies on the importance of Cdc25 and Ras-GTP for glucose-induced cAMP signalling [15,56^58].…”

“…Results obtained with cdc25 mutants can be explained by the fact that active Ras and therefore active Cdc25 is required for cAMP signalling by the Gpr1–Gpa2 system, but the signal does not run through Ras. Alternatively, inactivation of Cdc25 drastically reduces the amount of membrane‐bound adenylate cyclase (Pardo et al ., 1993). Deletion of the RAS genes also results in cytosolic localization of adenylate cyclase, but overexpression of Cdc25 in a ras1Δ ras2Δ bcy1Δ mutant again restores adenylate cyclase plasma membrane localization (Engelberg et al ., 1990).…”

“…Several distinct functional domains have been identi¢ed in this protein. Ras2p and the GEF, Cdc25p, associate with adenylate cyclase in this membrane-bound complex and the localisation thereof to the plasma membrane is dependent on Cdc25p [211]. The central leucine-rich repeat domain was identi¢ed as the Ras-responsive region of the protein [209] and also found to interact physically with Ras2p [210].…”
