Activation of Adenosine A2AReceptor Facilitates Brain-Derived Neurotrophic Factor Modulation of Synaptic Transmission in Hippocampal Slices

Abstract: It is concluded that presynaptic activity-dependent release of adenosine, through activation of A 2A receptors, facilitates BDNF modulation of synaptic transmission at hippocampal synapses.

“…As occurs with A 2A receptor agonists (Dió genes et al, 2004;Fontinha et al, 2008) and cAMP (Boulanger and Poo, 1999b), high-frequency neuronal firing triggers facilitatory actions of BDNF at excitatory synapses (Figurov et al, 1996;Boulanger and Poo, 1999b;Nagappan and Lu, 2005;Matsumoto et al, 2006). We therefore investigated whether TrkB receptors could be targeted to lipid rafts as a consequence of intense synaptic activity.…”

“…BDNF activates the TrkB tyrosine kinase receptor and the p75 receptor, which belongs to the tumor necrosis factor receptor family. We and others have reported that TrkB receptor function is modulated by activation of adenosine A 2A receptors (Lee and Chao, 2001;Dió genes et al, 2004;Mojsilovic-Petrovic et al, 2006;Fontinha et al, 2008;Tebano et al, 2008). This TrkB/A 2A receptor cross talk has two consequences, which may operate independently: (1) facilitation of BDNF-induced actions on synaptic transmission and plasticity by A 2A receptor agonists; and (2) direct phosphorylation and activation of TrkB receptors, in the absence of BDNF, a process called transactivation.…”

“…Slice preparation was carried out as previously described (Diógenes et al, 2004). Field excitatory postsynaptic potentials (fEPSPs) and whole-cell postsynaptic currents were recorded from the CA1 area of hippocampal slices (300-400 mm) upon stimulation of the Schaffer collateral fibers (rectangular pulses of 0.1-0.2 ms duration delivered each 15 s).…”

“…It is therefore feasible to speculate that the mechanism by which there is an impairment of the acquisition of conditioned responses by blockade of A 2A receptors could be a reduced activation of NMDA receptors. Another possibility is the interaction of adenosine A 2A receptors with TrkB receptors, as adenosine A 2A receptors, through a cyclic AMP-mediated process (Boulanger and Poo, 1999;Diógenes et al, 2004) or through transactivation of TrkB receptors (Lee and Chao, 2001), trigger BDNF actions at synapses. In this regard, it has been shown recently that TrkB receptors located in the hippocampal circuit are involved in associative learning and in learning-induced changes in synaptic strength (Gruart et al, 2007).…”
