Activation and turnover of the plant immune signaling kinase BIK1: a fine balance

Abstract: Mechanisms to sense and respond to pathogens have evolved in all species. The plant immune pathway is initiated by the activation of transmembrane receptor kinases that trigger phosphorylation relays resulting in cellular reprogramming. BOTRYTIS-INDUCED KINASE 1 (BIK1) is a direct substrate of multiple immune receptors in Arabidopsis thaliana and is a central regulator of plant immunity. Here, we review how BIK1 activity and protein stability are regulated by a dynamic interplay between phosphorylation and ubi… Show more

“…CPK28 phosphorylates the E3 ubiquitin ligases PLANT U-BOX 25 (PUB25) and PUB26, enhancing their ability to poly-ubiquitinate the RLCK BOTRYTIS INDUCED KINASE 1 (BIK1), a common substrate of multiple receptors and a critical signaling node in plant immunity (Monaghan et al, 2014;Wang et al, 2018;DeFalco & Zipfel, 2021). The CPK28-PUB25/26 regulatory module buffers BIK1 protein accumulation to optimize immune output (Gonçalves Dias et al, 2022). In the current study, we aimed to identify additional CPK28 binding partners in Arabidopsis using a proteomics approach.…”

“…In Arabidopsis thaliana (Arabidopsis), subfamily VII RLCKs (RLCK‐VIIs) BOTRYTIS‐INDUCED KINASE 1 (BIK1) and homologue PBS‐Like 1 (PBL1) are rapidly phosphorylated and activated by multiple PRR complexes to initiate intracellular signalling (Couto & Zipfel, 2016 ; Liang & Zhou, 2018 ). BIK1 associates with numerous immune signalling proteins including RESPIRATORY BURST OXIDASE HOMOLOGUE D (RBOHD), cyclic nucleotide gated ion channels CNGC2/CNGC4, and calcium‐dependent protein kinases (CDPKs) (Gonçalves Dias et al, 2022 ; Sun & Zhang, 2020 ). bik1 pbl1 loss‐of‐function plants display weakened defence responses and increased susceptibility to bacterial and fungal pathogens (Lu et al, 2010 ; Veronese et al, 2006 ; Zhang et al, 2010 ).…”
