Activation and thermostabilization effects of cyclic 2,3-diphosphoglycerate on enzymes from the hyperthermophilic Methanopyrus kandleri

Abstract: Enzymes involved in methane formation from carbon dioxide and dihydrogen in Methanopyrus kandleri require high concentrations (> 1 M) of lyotropic salts such as K2HPO4/KH2PO4 or (NH4)2SO4 for activity and for thermostability. The requirement correlates with high intracellular concentrations of cyclic 2,3-diphosphoglycerate (cDPG; approximately 1 M) in this hyperthermophilic organism. We report here on the effects of potassium cDPG on the activity and thermostability of the two methanogenic enzymes cyclohydrola… Show more

“…The role of DIP in the thermostabilization of enzymes remains questionable, although it has been shown to have a positive effect on the stabilization of glyceraldehyde-3-phosphate dehydrogenase of P. woesei, but did not increase the stability of hydrogenase and pyruvate ferredoxin oxidoreductase of Thermotoga maritima (Scholz et al, 1992;Ramakrishnan et al, 1997). The stabilizing effect of cBPG was also recently demonstrated on two enzymes involved in methane formation in Methanopyrus kandleri (Shima et al, 1998).…”

“…The last reaction in this synthesis is the only one exergonic enough to drive the synthesis up to cDPG rather than to its precursor 2,3-DPG. Also, at pH 7.0, cDPG is a trianion whereas 2,3-DPG is a penta-anion; thus cDPG has a smaller effect on the ionic strength than would 2,3-DPG (305).…”

“…Residues 174-175 are disordered. The coordinates were deposited in the RCSB Protein Data Bank 13 with accession number 2 3 7 M. The chemical identities of the two Zn 21 ions were established by an anomalous difference map. Data collection and refinement statistics are listed in Table I.…”
