Action of Aldosterone on Citrate Synthase in Cultured Renal Collecting Duct Cells

Abstract: Cultured renal collecting duct cells from neonatal rabbit kidney were used to examine the influence of aldosterone on enyzmatic activity of citrate synthase during increase in Na+ transport. Control epithelia showed citrate synthase activity of 71 ± 3 mU/mg protein (n=28), while after aldosterone treatment citrate synthase activity was significantly increased to 79 ± 6 mU/mgat 1 h(n=5), to 88 ± 6 mU/mg at 2 h(n=6) and to 93 ± 8 mU/mg protein at 3 h (n=5). Citrate synthase activity subsequently decre… Show more

“…Others have also found minimal or no increase in citrate synthase activity in cultured cells that respond to aldosterone by increasing Na' transport. Enzyme activity is not stimulated in either aldosterone-treated A6 cells or cultured toad bladder cells (33,34), and only a 30% rise has been demonstrated in primary cultures of rabbit CCD (35). These results in cultured cells stand in contrast to the marked stimulation seen in native tubules (32).…”

“…Cortical explants from the kidney of newborn New Zealand rabbits were isolated as described elsewhere [ 10, [23][24][25][26][27][28]. The explants consisted of a piece of capsula fibrosa with adherent nephrogenic blastema containing S-shaped bodies and collecting duct anlagen.…”

“…To overcome this problem, we developed a technique for culture of a clearly defined, highly differentiated, and electrically " tight" collecting duct epithelium. It consisted of "principal cells" only [lo, [23][24][25][26][27][28]. We used this system of cultured collecting duct cells from mammalian kidney to investigate the appearance of two collecting-duct-specific proteins (PcD 2 and 3) [7] in response to chronic aldosterone and AVP administration and in close correlation with electrophysiological properties.…”

“…[1][2][3][4][5] Adrenalectomy reduces renal citrate synthase activity, and the administration of aldosterone to adrenalectomized animals restores citrate synthase activity. 4 These findings arose from an ongoing effort to identify aldosterone-induced proteins (ie, proteins synthesized in response to the transcription factor activity of the occupied mineralocorticoid receptor) and to determine how these proteins mediate the classic action of mineralocorticoids: renal sodium reabsorption.…”
