The process of differentiation of digestive tract structures in the sterlet Acipenser ruthenus (L.) larvae was studied from hatching to the beginning of exogenous feeding [9 dph (day post hatching)] using histological procedures. On the day of hatching the digestive tract was closed and completely filled with nutrients (the yolk platelets) that were successively utilized during development. A liver primordium was present in the ventral region of the yolksac. The pancreas was observed on the 2 dph. At the same time, the mouth opening took place. Glandular and nonglandular stomach and anterior and intermediate intestine developed from the yolksac walls. Gastric glands became visible on the 7 dph. The primary intestine developed into the spiral intestine. At the moment of onset of exogenous feeding the yolk material was completely exhausted and there was not mixed feeding observed in sterlet larvae. The fish started exogenous feeding on the 9 dph, which was accompanied with evacuation of melanin plug. At the end of endogenous feeding the digestive tract of sterlet larvae was developed and functional, so they could properly utilize food.

This study describes the stomach development in the sturgeon Acipenser naccarii, focusing on its histoenzymatic and ultrastructural characteristics from hatching until the end of the juvenile phase, 1 month later. The study aims to assist the aquaculturist in identifying the best timing for the start of feeding and to select the appropriate food for each stage of A. naccarii development. At hatching, the anterior portion of the digestive system consists of a yolk sac with a large central yolk mass. The yolk mass is enclosed by differentiating endodermal epithelium. The most significant ultrastructural characteristics of the epithelium are the numerous supranuclear vacuoles containing protein and lipid yolk granules, dilated intercellular spaces, and filiform cytoplasmic projections related to the uptake of reserve material on the apical surface. Newly hatched eleutheroembryos show optimal enzymatic activities at a pH of 1.5 and 3, which is probably related to the presence of hatching glands and the digestion of yolk components. During the free-embryo phase, the yolk sac epithelium undergoes a series of histological, ultrastructural, and functional modifications to form a cecal stomach with anatomical characteristics similar to those of juvenile specimens. Gastric glands emerge between 6 and 7 days post-hatching (PH), although the absence of secretion granules in their oxyntic-peptic cells indicates that the stomach is not yet functional. During the following days (the exotrophic period), development of the tubular-vesicular network is observed, and secreting granules occur in the cytoplasm of oxyntic-peptic cells until around day 35 after fertilization (day 27 PH), when acid protease enzymatic activity is again detected. These histological and enzymatic changes and the greater number and size of oxyntic-peptic cell secretion granules appear to be related to the gastric compartment ontogeny reaching functionality at this developmental stage.

This study characterized the ultrastructure of normal epidermal and gastrointestinal epithelial cells of larval hybrid striped bass (white bass Morone chrysops × striped bass M. saxatilis) that were fed live prey (brine shrimp Artemia spp. nauplii) or an artificial dry diet. Scanning electron microscopy of larvae that were given live feed revealed normal epidermal cells with microridge structures and proliferation of neuromasts along the lateral line. Cell junctions had double microridge structures. Transmission electron microscopy of the gut showed keratinized epithelial cells with underlying mucous cells in the foregut, while the midgut was characterized by columnar epithelial cells with extensive pinocytotic activity. The hindgut had columnar epithelial cells with centrally located nuclei, regularly spaced microvilli, and numerous tubular mitochondria surrounded by rough endoplasmic reticulum. Larvae that received live feed reached metamorphosis by 27 d posthatch (dph), and clearly identifiable gastric glands were present. Larvae that were given the artificial diet did not develop; both epidermal and gastrointestinal cells showed apparent osmotic stress by 10 dph, and the condition worsened through 24 dph. Stress was indicated by loss of microridges in epidermal cells and eventual necrosis. Increased keratinization and reduced pinocytotic activity were observed in midgut cells, whereas hindgut epithelial cells showed dissociation of the rough endoplasmic reticulum and reduced numbers of mitochondria. However, tight cell junctions, desmosomes, and double microridges at cell junctions persisted.
