Acinetobacter baumannii outer membrane protein a modulates the biogenesis of outer membrane vesicles

Abstract: Acinetobacter baumannii secretes outer membrane vesicles (OMVs) during both in vitro and in vivo growth, but the biogenesis mechanism by which A. baumannii produces OMVs remains undefined. Outer membrane protein A of A. baumannii (AbOmpA) is a major protein in the outer membrane and the C-terminus of AbOmpA interacts with diaminopimelate of peptidoglycan. This study investigated the role of AbOmpA in the biogenesis of A. baumannii OMVs. Quantitative and qualitative approaches were used to analyze OMV biogenesi… Show more

“…It appears that OmpA-like domain links the outer membrane to the peptidoglycan layer and is essential for the integrity of the cell. Previous quantitative and qualitative approaches have shown that outer membrane vesicles (OMVs) production is significantly increased in the ΔAbOmpA mutant compared to the wild-type bacteria [34]. This data confirms that OmpA-like domain of AbOmpA plays an important role in the integrity of the outer membrane which may give rise to a controlled shedding of OMV from the surface of the bacteria.…”

“…2C). OmpA is an outer membrane protein that is commonly found in Acinetobacter species (59), and IEM with anti-OmpA antibodies attached to gold particles confirmed the presence of OmpA in the OMVs (Fig. 2B).…”

“…and Acinetobacter baumannii mutants lacking OmpA display increased OMV production 34–37 , most probably as a consequence of decreased crosslinking between PG and the outer membrane 30 . Interestingly, although modulation of this type of envelope crosslink also seemed to lead to hypervesiculation in Pseudomonas aeruginosa mutants lacking OprF (an OmpA homologue 38 ), in this case hypervesiculation is actually a consequence of an increase in the levels of Pseudomonas quinolone signal (PQS), which stimulates OMV production in P. aeruginosa 39 (see below).…”
