Acheron/Larp6 Is a Survival Protein That Protects Skeletal Muscle From Programmed Cell Death During Development

Abstract: The term programmed cell death (PCD) was coined in 1965 to describe the loss of the intersegmental muscles (ISMs) of moths at the end of metamorphosis. While it was subsequently demonstrated that this hormonally controlled death requires de novo gene expression, the signal transduction pathway that couples hormone action to cell death is largely unknown. Using the ISMs from the tobacco hawkmoth Manduca sexta, we have found that Acheron/LARP6 mRNA is induced ∼1,000-fold on the day the muscles become committed t… Show more

“…LARP6 , one of the co-expressed genes associated with the lncRNA G8399 encodes a protein that prevents cell death until it is triggered by dephosphorylation and degraded ( Sheel et al, 2020 ). We also found that EOMES, one of the target genes for the lncRNA G8399, is involved in the trophoblast differentiation as well as gastrulation, regulating both the mesodermal delamination, endodermal specification, which are essential for developing the tissues extraembryonic to the uterus during the mammalian embryogenesis ( Kidder and Palmer, 2010 ; Probst and Arnold, 2017 ).…”

“…Likewise, the interconnection between apoptosis and mitophagy shows interactions between both protagonists and even a dual role for some proteins [1]. In mammals, BNIP3 is associated with a pro-apoptotic function [24], whereas, in Drosophila, few studies have focused on the role of BNIP3 in mitophagy [25][26][27], and, to our knowledge, only one explored the role of BNIP3 in apoptosis [28]. Concerning PINK1, Han et al observed increased levels of apoptosis induced by a mutation altering the PINK1 kinase activity in Drosophila [29].…”

“…The ISMs are a classical model system for skeletal muscle atrophy and death ( Schwartz and Truman 1982 ; Schwartz 2018 ; Sheel et al, 2020 ). These muscles are composed of giant syncytial cells, each of which is about 5 mm long and up to 1 mm in diameter.…”
