Acetyl-11-Keto-β-Boswellic Acid (AKBA) Prevents Lipopolysaccharide-Induced Inflammation and Cytotoxicity on H9C2 Cells

Abstract: Acetyl-11-keto-beta-boswellic acid (AKBA), the major component of Boswellia serrata, exhibits anti-inflammatory activities. This in vitro study investigated the protective effects of AKBA against lipopolysaccharide (LPS)-induced cardiac dysfunction. In this study, the H9C2 cardiomyocytes were pretreated with AKBA (2.5, 5, and 10 μM for 24 h), and then cotreated with LPS for another 24 h. The MTT assay, ELISA test kits, and quantitative real-time PCR analysis assessed the cell viability, levels of proinflammato… Show more

“…H9c2 rat cardiomyoblast cells were utilized to investigate the protective effects of carnosol (5, 10, and 20 μ M) against inflammation resulting through LPS for 24 h, then followed cotreatment with LPS (10 μ g/mL) stimulation [ 23 ]. H9c2 cells were routinely cultured in RPMI-1640 supplemented with 10% (v/v) FBS and 1% (v/v) penicillin/streptomycin, which was incubated with 5% v/v CO 2 at 37°C.…”

“…After the cell treatment in the first and second experiments (at a density of 2 × 10 6 cell/well), the supernatant of cells was gathered to measure the secretory levels of inflammatory mediators TNF- α , IL-1 β , IL-6, and PGE 2 , according to the manufacturer's protocol with the help of the corresponding ELISA kits [ 23 , 24 ].…”

“…More recently, according to in vitro LPS-induced inflammation on H9C2 cells, an in vivo zebrafish larval model and molecular docking study, Boswellia extract and BAs inhibited inflammation and cytotoxicity and showed strong redox activity. The mechanism of action may be by increasing anti-inflammatory activity by decreasing specific inflammatory gene expression (iNOS, TNF-α, IL-1, and COX-2) (Siddhu et al 2022 ; Taherzadeh et al 2022 ). Taken together, the results of the above studies show evidence that GA and BA suppress SARS-CoV-2-induced inflammation through inhibition of the NLRP3 inflammasome pathway and HMGB1 release.…”

“…3-O-acetyl-11-keto-β-boswellic acid (AKBA) is the major boswellic acid isolated from B. serrata [ 28 ]. Accumulating experimental and clinical data attest to the exceptional ability of AKBA to treat several disorders, including infection, cancer, sepsis, hypertension, arthritis, liver and brain damage, memory deficits, and colitis due to its potent antioxidant and anti-inflammatory effects [ 28 , 29 , 30 , 31 , 32 , 33 ]. In addition, studies have shown that AKBA can act on several cellular targets, including transcription and growth factors, as well as signaling mediators and pathways [ 28 ].…”
