Over the past few decades, gellan gum (GG) has attracted substantial research interest in several fields including biomedical and clinical applications. The GG has highly versatile properties like easy bio-fabrication, tunable mechanical, cell adhesion, biocompatibility, biodegradability, drug delivery, and is easy to functionalize. These properties have put forth GG as a promising material in tissue engineering and regenerative medicine fields. Nevertheless, GG alone has poor mechanical strength, stability, and a high gelling temperature in physiological conditions. However, GG physiochemical properties can be enhanced by blending them with other polymers like chitosan, agar, sodium alginate, starch, cellulose, pullulan, polyvinyl chloride, xanthan gum, and other nanomaterials, like gold, silver, or composites. In this review article, we discuss the comprehensive overview and different strategies for the preparation of GG based biomaterial, hydrogels, and scaffolds for drug delivery, wound healing, antimicrobial activity, and cell adhesion. In addition, we have given special attention to tissue engineering applications of GG, which can be combined with another natural, synthetic polymers and nanoparticles, and other composites materials. Overall, this review article clearly presents a summary of the recent advances in research studies on GG for different biomedical applications.

To elaborate the decisive role of surfactants in promotion of aceclofenac' skin absorption, potentially avoiding irritation, we developed non-ionic microemulsions varying natural or synthetic surfactants: sucrose esters (laurate or myristate) vs. polysorbate 80. A comprehensive physicochemical characterization indicated no significant influence of the solubilized nonsteroidal anti-inflammatory drug on the bicontinuous structure of blank formulations. To evaluate skin tolerability of isopropyl alcohol, a sucrose ester-based microemulsion containing transcutol P as a cosurfactant was also developed. The measured skin parameters strongly depended on the (co)surfactant type, showing higher compatibility of the microemulsions containing sucrose ester and isopropyl alcohol. In vitro release results, in vivo tape stripping and pharmacokinetics in rats confirmed superiority of the sucrose ester- over polysorbate-based microemulsions (total amounts of aceclofenac penetrated 60.81±5.97 and 60.86±3.67 vs. 27.00±5.09μg/cm(2), and its maximum plasma concentrations 275.57±109.49 and 281.31±76.76 vs. 150.23±69.74ng/ml for sucrose laurate- and myristate- vs. polysorbate 80-based microemulsions, respectively). Hence, sugar-based excipients increased delivery of aceclofenac through stratum corneum by increasing its fluidity, showing overall more satisfying safety profiles. In conclusion, sucrose ester-based microemulsions proved to be promising carriers for dermal/transdermal aceclofenac delivery.

In the present work, in view of the medicinal properties of vetiver oil (extracted from the roots of Vetiveria zizanioides L.), we made an attempt to encapsulate vetiver oil in a biocompatible polymeric system made of sodium alginate with gellan gum or karaya gum. Sodium alginate and gellan gum or karaya gum were ionotropically cross-linked to encapsulate vetiver oil. Vetiver oil encapsulations in these microcapsules were 35.92 ± 3.18 % to 78.55 ± 3.35%. Vetiver oil encapsulated microcapsules were of spherically shaped with 656-769 µm mean diameter. This vetiver oil encapsulated microcapsules made of alginate-gellan gum blends were found capable of providing a long release of encapsulated oil, showing the potential for the sustained release application. These microcapsules were analyzed by FTIR, DSC, and SEM, etc. In addition, sedative and hypnotic activities of vetiver oil encapsulated polymeric microcapsules in the male Swiss albino mice were evaluated. The sedative-hypnotic activity of vetiver oil encapsulated polymeric microcapsules in rats was observing the number of crossing and motilities. The results proves that vetiver oil encapsulated polymeric microcapsules decreased motility when compared to the control group.
