Accurate identification of Australian mosquitoes using protein profiling

Abstract: Australian mosquito species significantly impact human health through nuisance biting and the transmission of endemic and exotic pathogens. Surveillance programmes designed to provide an early warning of mosquito-borne disease risk require reliable identification of mosquitoes. This study aimed to investigate the viability of Matrix-Assisted Laser Desorption/Ionization–Time-of-Flight Mass Spectrometry (MALDI-TOF MS) as a rapid and inexpensive approach to the identification of Australian mosquitoes and was vali… Show more

“…pipiens complex using multiplex PCR method in itself. There has been some research undertaken using DNA barcoding [47] and protein profiling [48] to distinguish the subspecies of the complex mentioned above. Since the adopted primers in this study only focused on three forward primers (ACEquin, ACEpall, and ACEpip) and one backward primer (B1246s), plus the limitation of sampling to some extent, Cx.…”

“…have also highlighted the need for sharing and have expressed interest in an international database (Raharimalala et al ., 2017). Many reference spectra are also available on request or directly online (Lawrence et al ., 2019), substantially facilitating database creation. The MALDI-TOF mass spectrometry approach may also appear to be limited by the cost of the device but when the MALDI-TOF MS device is bought for clinical microbiology purposes, it can also be used for medical entomology at no additional cost.…”

“…MSPs were created on the basis of an algorithm using peak position, intensity and frequency data. Between two and nine new reference spectra per species were added to the lice database in our laboratory [26].…”

“…Recently, MALDI-TOF MS has been evaluated as an efficient tool for the identification of arthropods including ticks [2,5], mosquitoes [26,28,46], culicoides [6,35], fleas [30,48], triatomines [25], tsetse flies [15], and phlebotomines [9] in laboratory and field conditions.…”
