Accurate Detection of Methicillin-Resistant Staphylococcus aureus in Mixtures by Use of Single-Bacterium Duplex Droplet Digital PCR

Abstract: Accurate and rapid identification of methicillin-resistant (MRSA) is needed to screen MRSA carriers and improve treatment. The current widely used duplex PCR methods are not able to differentiate MRSA from coexisting methicillin-susceptible (MSSA) or other methicillin-resistant staphylococci. In this study, we aimed to develop a direct method for accurate and rapid detection of MRSA in clinical samples from open environments, such as nasal swabs. The new molecular assay is based on detecting the cooccurrence o… Show more

“…The study by Rolo et al ( 2017 ) provided support that the Staphylococcal Chromosomal cassette (SCC mec ), a mobile genetic element, evolved from three Staphylococcus species— S. vitulinus, S. fleuretti , and S. sciuri . Since its original development, the antibiotic resistance gene remains highly conserved in chromosomes of MRSA, and is currently used as a marker for screening and identifying MRSA isolates (Koupahi et al, 2016 ; Luo et al, 2017 ). Up to the present, 11 types of SCC mec (40-60Kb) have been identified (IWG-SCC, 1999 ).…”

“…A great number of studies use dPCR to diagnose infectious disease-related viruses, including hepatitis B virus (HBV), cytomegalovirus, human influenza, and HIV [26][27][28]; bacterial infections (Mycobacterium tuberculosis, Chlamydia trachomatis, and Staphylococcus aureus) [29][30][31]; and parasitic infectious such as detection of Plasmodium falciparum and Plasmodium vivax [32].…”

“…Some of the technologies are worth mentioning separately given the fact that they can be considered extremely elegant from an experimental design point of view. Digital droplet PCR for instance was shown to be sensitive and rapid and it has to be realized that instruments allowing in house development of droplet-based PCR tests are already (commercially) available ( Luo et al, 2017 ). Nanowires are attractive because of size, relatively low costs, and speed of the assay and broad applicability of the technology which, in addition, is easy to multiplex ( Ibarlucea et al, 2017 ).…”
