Accumulation of Poliovirus Proteins in the Host Cell Nucleus

Abstract: Poliovirus (type 1, Mahoney) proteins were transported into the nuclei of HEp-2 cells, as demonstrated by means of electron microscopic autoradiography. Quantitative determinations of the proteins by electrophoresis showed that the relative amounts of precursor proteins (la, 3b, lb), of some of the intermediate polypeptides (3a, 3c, 5b, 2), and of one end product (VP1) were higher in the nucleus than in the cytoplasm. Other proteins (VPO, YP2, X, 4, 6a) were found in the same relative amounts in the nucleus as… Show more

“…Host RNA synthesis inhibition by picornaviruses is induced by viral proteins [8]; the inhibiting protein(s), however, have never been iden tified. Specific poliovirus proteins were shown to accumulate in the host cell nuclei [9,10], …”

“…Thus, viral proteases 2A and 3C were reported to target several transcription factors (56,73,(78)(79)(80) and histones (21). Accumulation of virus-specific proteins in the nuclei of poliovirus-infected (8,22) and encephalomyocarditis virus-infected (3, 4) cells was directly observed. Nuclear alterations developing upon different forms of picornavirus-induced apoptosis (7,28,35,40,50,69) also require entry of certain proapoptotic host proteins (e.g., effector caspases and DNases) into the nuclei (cf., references 20 and 55).…”

“…This fact, however, does not mean that the nuclei are not involved in the infectious process. Indeed, virusspecific proteins have been detected in the nuclei of poliovirusinfected (11,29) and encephalomyocarditis virus (EMCV)-infected (5, 6) cells. Poliovirus proteases 2A and 3C are known to target a variety of nuclear transcription factors and histones (66,78,79,80).…”
