Accumulation of NEDD8 in neuronal and glial inclusions of neurodegenerative disorders

Abstract: NEDD8 (neural precursor cell expressed, developmentally down-regulated 8) is a ubiquitin-like protein that controls vital biological events through its conjugation to members of the cullin family, which are components of certain ubiquitin E3 ligases. Recent studies have shown that NEDD8 is incorporated into Lewy bodies (LBs) in Parkinson's disease, Mallory bodies in alcoholic liver disease and Rosenthal fibres in astrocytoma. In order to examine whether NEDD8 plays a role in the formation of ubiquitinated incl… Show more

“…The condition in these cells could lead to the depletion of free Ub, which might trigger the synthesis of the heterologous chains. Indeed, Nedd8 has been shown to be present in protein aggregates in these neurodegenerative diseases (113). The formation of the heterologous chains replicates the function of Ub chains and at the same time ensures avoidance of the further depletion of free Ub within cells.…”

“…In normal or physiological condition, synphilin-1 is predominantly expressed in neurons, located in the cytoplasm and presynaptic nerve terminals, and associated with synaptic vesicles. 8,28 However, in several neurodegenerative disorders called ␣-synucleinopathies, such as PD, DLB, and MSA, synphilin-1 is mainly localized in neuronal and glial cytoplasmic inclusions, 10,11 in which ␣-synuclein, 32,34 ubiquitin, 35 NEDD8, 14,36 and the proteasome 25,37 are also present. Experimental studies have revealed that the co-expression of synphilin-1 with ␣-synuclein results in the formation of LB-like cytoplasmic inclusions in cultured HEK293 cells.…”

“…13 Recently, NEDD8 has been shown to be incorporated into a variety of ubiquitinated inclusions, including the neurofibrillary tangles present in AD. 14 APP is the precursor of the amyloid b (Ab) peptide, the main component of senile plaques in AD. APP is synthesized as a transmembrane glycoprotein composed of extracellular and cytoplasmic domains.…”
