Lead (Pb) is a hazardous pollutant in water environments that can cause significant damage to aquatic animals and humans. In this study, crucian carp (Carassius auratus) were exposed to waterborne Pb for 96 h; then, histopathological analysis, quantitative qPCR analysis, and 16S high-throughput sequencing were performed to explore the effects of Pb on intestinal bioaccumulation, structural damage, oxidative stress, immune response, and microbiota imbalance of C. auratus. After Pb exposure, the intestinal morphology was obviously damaged, including significantly increasing the thickness of the intestinal wall and the number of goblet cells and reducing the depth of intestinal crypts. Pb exposure reduced the mRNA expressions of Claudin-7 and villin-1 while significantly elevated the level of GST, GSH, CAT, IL-8, IL-10, IL-1, and TNF-α. Furthermore, 16S rRNA analysis showed that the Shannon and Simpson indices decreased at 48 h after Pb exposure, and the abundance of pathogenic bacteria (Erysipelotrichaceae, Weeksellaceae, and Vibrionaceae) increased after Pb exposure. In addition, the correlation network analysis found that Proteobacteria were negatively correlated with Firmicutes and positively correlated with Bacteroidetes. Functional prediction analysis of bacteria speculated that the change in intestinal microbiota led to the PPAR signaling pathway and peroxisome function of the intestine of crucian carp was increased, while the immune system and membrane transport function were decreased. Finally, canonical correlation analysis (CCA) found that there were correlations between the intestinal microbiota, morphology, antioxidant factors, and immune factors of crucian carp after Pb exposure. Taken together, our results demonstrated that intestinal flora dysbiosis, morphological disruption, oxidative stress, and immune injury are involved in the toxic damage of Pb exposure to the intestinal structure and function of crucian carp. Meanwhile, Pb exposure rapidly increased the abundance of pathogenic bacteria, leading to intestinal disorders, further aggravating the damage of Pb to intestinal structure and function. These findings provide us a basis for the link between gut microbiome changes and heavy metal toxicity, and gut microbiota can be used as biomarkers for the evaluation of heavy metal pollution in future.

Sea cucumbers Apostichopus japonicus were exposed to five concentrations of dietary hexavalent chromium [0 (control), 100, 200, 400, and 800 mg Cr6+/kg dry weight] amended with K2Cr2O7 for 30 days. The bioaccumulation and immune responses [antioxidant enzymes: superoxide dismutase (SOD) and catalase (CAT); hydrolytic enzymes: acid phosphatase (ACP) and alkaline phosphatase (AKP)] of sea cucumbers were subsequently evaluated. This study found that the order of Cr accumulation in the experimental tissues was respiratory tree > intestine > body wall. Significantly lower SOD activities occurred in the 400 mg/kg group compared to that in the control group. Higher dietary Cr6+ exposure (400 and 800 mg Cr6+ /kg dry weight) did not negatively alter the CAT activities, but significantly inhibited CAT activities in 100 mg/kg group, compared to control group. ACP activities in groups 200, 400 and 800 mg/kg were significantly lower than those in control group, while no significant differences occurred in AKP activities among groups. The present study provides important information into the bioaccumulation and immune responses of the sea cucumber A. japonicus in response to chronic dietary Cr6+ exposure.

Leather tanning mainly depends on chrome tanning. A huge amount of basic chromium sulfate (BCS) is used in leather tanning. About 60–70% of BCS has been taken by leather and the rest of the amount is discharged as effluent and solid waste. In the Common Effluent Treatment Plant (CETP) of BSCIC Tannery Industrial Estate (Hemayetpur, Savar, Dhaka), the effluent which contained a high concentration of chromium is precipitated as solid chromium hydroxide with other elements which is called a chrome cake. Chrome cake is not exposed directly in the open space due to its toxic effect on human health as well as the environment. Human occupational experience distinctly indicates that, when inhaled, chromium compounds are respiratory tract irritants, resulting in airway irritation, airway obstruction, and lung, nasal, or sinus cancer. It is strongly suggested to extract Cr from the chrome cake before dumping it; otherwise, it will be a potential hazard to the environment. Herein, the chrome cake is characterized by inductively coupled plasma-optical emission spectroscopy (ICP-OES), X-ray fluorescence (XRF), UV-visible spectrophotometer, etc., and develops a method to separate chromium from chrome cake. ICP-OES and XRF analyses show that the total chromium contained in chrome cake is ∼17.45% and 20.7%, respectively. UV-visible spectrophotometer indicates that ∼0.04% of chromium is hexavalent. Afterward, chromium has been separated by using H2SO4 where the optimized concentration was found to be 7%. The separation efficiency of chromium from the chrome cake for optimized condition is obtained at ∼95%.
