Acceptability of Mini-Tablets in Young Children: Results from Three Prospective Cross-over Studies

Abstract: To ensure optimal, reliable treatment, it is necessary to investigate the efficacy, safety and the optimal dose of drug substances and to develop suitable age-specific pharmaceutical formulations for the different paediatric age groups due to a lack of evidence-based therapeutic options for children. While WHO recommends the use of solid dosage forms in general, European Medicines Agency (EMA) requires evidence for the suitability of these dosage forms in the targeted age group. This review aims to summarize a… Show more

“…Despite the promise hold by multi-particulate formulations these dosage forms are not yet a well-established platform mainly due to manufacture and administration constrains, such as weight control, content uniformity and handling by patients. The impact on adherence has not been yet studied even though higher acceptability and capacity to swallow mini-tablets over syrup has been demonstrated in the paediatric population [102,103].…”

“…The various types of marketed oral dosage forms include solutions, syrups, suspensions, powders, granules, tablets, orally disintegrating tablets (ODT), chewable tablets, dispersible tablets, capsules, oral thin strips, chewing gums, powder for reconstitution, tablets for constituton to suspension, oral drops and troches [10] . Chewable tablets, orodispersible formulations such as ODT, orally disintegrating mini-tablets and orodispersible films (ODF) and multi-particulate formulations can be conveniently consumed by school going children and geriatric patients due to advantages like convenient administration and less swallowing difficulties [11][12][13][14][15] . A flexible tablet formulation administered as chewable tablets, ODT, orally dispersible tablets dispersed in a small quantity of water, and also be orally swallowed.…”

“…Recently, the miniaturization of tablets has emerged as a focus of pharmaceutical research. 1,2) Mini-tablets, defined by WHO as tablets with a diameter of ≤4 mm, 3) have attracted attention as a new dosage form to improve adherence, and their pharmaceutical and clinical suitability has been investigated. 4,5) Many pharmaceutical aspects have been studied, including the development of mini-tablets with a diameter of 1-2 mm to increase miniaturization, enhance dissolution control, and the improvement of absorption by modification of the tablet surfaces.…”
