Abstract 4311: Fluorescence-guided surgery of pancreatic cancer using activatable cell penetrating peptides (ACPPs) in orthotopic mouse models

Abstract: Surgical resection with complete removal of tumor remains one of the most effective modalities in the treatment of cancer. The aim of this study was to evaluate the efficacy of using activatable cell penetrating peptides (ACPPs) conjugated to Cy5 fluorophores to accurately label primary pancreatic tumor for fluorescence-guided surgery (FGS) in orthotopic mouse models. Orthotopic mouse models of human pancreatic cancer using surgical implantation of tumor fragments of the MiaPaca-2-GFP pancreatic cancer cell li… Show more

“…Using indocyanine green, Yuen and co-workers have demonstrated detection of sentinel lymph nodes as well as lymphatic vessels during open prostatectomy . Metildi and co-workers have reported use of activatable cell penetrating peptides (ACPPs) in fluorescence-guided surgery of pancreatic cancer using orthotopic mouse models . Yano et al have developed a telomerase-specific oncolytic adenovirus (OBP-401) that expresses green fluorescent protein (GFP) and have used this GFP-expressing adenovirus to label pancreatic cancer for fluorescence-guided surgery in a mice tumor model …”

“…After the cleavage of the MMP-responsive linker in tumors, the inhibitory polyanionic domains diffused and CPPs could be thus unleashed and take its cargo into cells. Since then, this strategy has quickly aroused researchers' attention and ACPP-based drug delivery systems have been thoroughly investigated and applied to tumor imaging or tumor management [83][84][85][86][87][88][89][90][91][92]. This measure could even be adopted in fluorescence-guided surgery in tumor models [90,91].…”

This measure could even be adopted in fluorescence-guided surgery in tumor models. , Savariar et al discovered a family of novel ratiometric activatable cell penetrating peptides, which contained Cy5 as far red fluorescent donor and Cy7 as near-infrared fluorescent acceptor . Cy5 was quenched by the fluorescence resonance energy transfer (FRET) with Cy7.…”
