Absorption of Nasal and Bronchial Fluids: Precision Sampling of the Human Respiratory Mucosa and Laboratory Processing of Samples

Abstract: The methods of nasal absorption (NA) and bronchial absorption (BA) use synthetic absorptive matrices (SAM) to absorb the mucosal lining fluid (MLF) of the human respiratory tract. NA is a non-invasive technique which absorbs fluid from the inferior turbinate, and causes minimal discomfort. NA has yielded reproducible results with the ability to frequently repeat sampling of the upper airway. By comparison, alternative methods of sampling the respiratory mucosa, such as nasopharyngeal aspiration (NPA) and conve… Show more

“…Reasons for exclusion are described in detail in Figure E2, the commonest being lack of ICS therapy. [20][21][22][23][24] (ACT 25 point scale -score 20-24 well controlled, less than 20 not well controlled).…”

“…Concentrated nasal lining fluid was collected by applying synthetic absorptive matrices to the nasal mucosa for 1 minute and stored at -80 o C until analysis 21 . Fluid was eluted by adding 100µL Luminex assay buffer, supernatant was clarified by centrifugation for 10 minutes, and 30 cytokines and immune proteins were analysed as per manufacturer's protocol as we previously described 9,19 .…”

“…Data from these four devices and an embedded asthma symptom diary will be recorded in the STIFLE application through an integrated approach (figure 3). ∼30 patients who agree to participate in a nasal biomarker substudy of STIFLE will be shown how to collect a nasal absorption sample (synthetic absorptive matrix (SAM) [28]), and will be provided with a kit and instructions for collecting the samples at home. These patients will collect daily morning nasal samples for the first 30 days of randomised treatment for the evaluation of inflammatory biomarkers.…”
