Abscisic Acid and Cytokinin Contents of Leaves in Relation to Salinity and Relative Humidity

Abstract: The question is raised whether the hormonal modifications in a plant exposed to osmotic root stress result directly from the decrease in water potential of the root environment or from disturbances of the plant's water balance.Tobacco plants were held for 24 hours under either high or low relative humidities, with or without salt.

“…Water and osmotic stresses alter the content of endogenous cytokinin and ABA: cytokinin activity in the root exudate declines drastically after 48 hr of salinity stress (!tai, Richmond, and Vaadia 1968), and ABA content in leaves of such plants increases considerably (Wright and Hiron 1969;Mizrahi et al 1971) the increase starting within 40 min of commencement of salinity stress (Mizrahi and Richmond, unpublished data). This study describes the effects of ABA and kinetin on plant response to various conditions which impair plant water balance.…”

“…It has been implicated in a variety of physiological processes (1,11,13,15) and is found in elevated levels under several stressful conditions (3,4,7,8,16,18,23 Samples for ABA determination were harvested at 0, 12, 24, 48, and 72 h. Zero sampling time (09:00 h) was 3 h after the lights came on; therefore, all samples, except those at 12 h, were harvested 3 h after the lights came on. The 12-h samples were taken 1 h before the end of the light period (2 1:00 h).…”

“…Gas chromatography on a Packard model 873 gas chromatograph equipped with a model 873 electron capture detector was carried out as detailed by Mizrachi et al (1971). Peaks indicating endogenous ABA were interpolated from identical retention times of marker ABA.…”

“…The extraction procedure was that of Mizrachi et al (1971) and the final extract was chromatographed as described above. Commercial ABA (Sigma-mixed isomers) was used as a marker on the chromatograms.…”
