Ablation of NF1 function in neurons induces abnormal development of cerebral cortex and reactive gliosis in the brain

Abstract: Neurofibromatosis type 1 (NF1) is a prevalent genetic disorder that affects growth properties of neural-crest-derived cell populations. In addition, approximately one-half of NF1 patients exhibit learning disabilities. To characterize NF1 function both in vitro and in vivo, we circumvent the embryonic lethality of NF1 null mouse embryos by generating a conditional mutation in the NF1 gene using Cre/loxP technology. Introduction of a Synapsin I promoter driven Cre transgenic mouse strain into the conditional NF… Show more

“…Nf1 +/− and WT mice, as well as Dlx-Cre + ;Nf1 flox/+ and their control littermates, were on a hybrid background of C57BL/6NTac and 129T2/ SvEmsJ. Dlx-Cre + ;Nf1 flox/+ mice and controls were from a cross between a line expressing Cre under the Dlx promoter and a line expressing a floxed Nf1 allele (46). All experiments used littermates as controls and were done with the experimenter blinded to genotype.…”

“…Nf1 flox/flox , Tsc1 flox/flox , and Pten flox/flox mice were generated as previously described (16)(17)(18), whereas Nf1 +/− ; LSL-Rheb; GFAP-Cre and Nf1 +/− ; Tsc1 flox/flox ; GFAP-Cre mice were generated by successive intercrossing (16,22,41) with age-matched mice as controls. GFAP-Cre and LSL-Rheb littermates were indistinguishable from C57BL/6 mice.…”
