Aberrant Notch signaling in gliomas: a potential landscape of actionable converging targets for combination approach in therapies resistance

Abstract: The current therapeutic protocols and prognosis of gliomas still depend on clinicopathologic and radiographic characteristics. For high-grade gliomas, the standard of care is resection followed by radiotherapy plus temozolomide chemotherapy. However, treatment resistance develops due to different mechanisms, among which is the dynamic interplay between the tumor and its microenvironment. Different signaling pathways cause the proliferation of so-called glioma stem cells, a minor cancer cell population with ste… Show more

“…In turn, high total NOTCH1 immunostaining is associated with progression of kidney renal clear cell carcinoma contradicting TCGA prediction (103,104). Likewise, in contrast to TCGA data, literature suggests association of NOTCH1 expression and glioma progression by modulating CXCL12/CXCR4 (105,124).…”

“…The leading mechanism of resistance in glioma cells is the high activity of O-6-methylguanine DNA methyltransferase (MGMT), which repairs TMZ-induced DNA damage and contributes to TMZ resistance [ 100 ]. TMZ resistance in GBM has also been linked to a number of cellular signalling pathways, including Hedgehog (HH) [ 101 ], NF-κB [ 102 ], Wnt/β-catenin (Wnt) [ 103 ], and Notch [ 104 ].…”

“…Furthermore, mutations in the WNT pathway’s key components dramatically regulate the WNT pathway. According to one study, the miR-138-2-3p and miR-770-5p are widely expressed in GBM and play an important role in Wnt signaling abnormalities in GBM, but they can also affect -catenin expression in cancers such as hepatocellular carcinoma and laryngeal tumors [ 112 ].…”
