Aberrant methylation of EDNRB and p16 genes in hepatocellular carcinoma (HCC) in Taiwan

Abstract: Epigenetic alternation via the promoter hypermethylation of putative tumor suppressor genes has been implicated in the development of hepatocellular carcinoma (HCC). In this study, we investigated the epigenetic changes in two candidate tumor suppressor genes, endothelin receptor type B (EDNRB) and p16, and their relation to the expression of these two genes in HCC. Methylation-specific polymerase chain reaction (MS-PCR) was performed to analyze the promoter methylation status of the EDNRB and p16 genes in tum… Show more

“…This was found also in colorectal cancer cells and cancer associated cells (fibroblasts and endothelial cells) in comparison with normal tissue (17). In line with these findings in colorectal cancer, there is similar expression in other gastrointestinal cancers such as gastric cancer (19,36) and hepatocellular cancer (18). Regarding the colorectal cancer cell lines LIM1215, HT29, SKCO1, SKCO17 and LoVo, there is high expression of EDNRA and low expression of EDNRB in comparison to normal colon (35).…”

“…Endothelin receptors type B is considered as the predominant receptor in normal colon, but in colon cancer it is down-regulated in cancer associated blood vessels, fibroblasts and epithelial cells (17). Moreover, epigenetic alteration of EDNRB expression by aberrant methylation has an important role in the pathogenesis of hepatocellular carcinoma (18), and in gastric cancer (19).…”

“…These data suggest that loss of tumor suppressor gene in 13q may be an early event in the Table 4). Alterations of EDNRB are frequent in a wide range of malignancies, in leiomyosarcoma as well as in other soft-tissue sarcomas (Hsu et al 2006). Array-CGH method has revealed recurrent losses at10q in the region of 10q23.31, 10q 25.2-q25.3, and 10q26.2.…”
