Aberrant expression of cell‐free nucleosomes in dermatomyositis/polymyositis

Abstract: Nucleosomes are typically located intracellularly, and extracellular levels of nucleosomes indicated the degree of cell death. We postulated aberrant nucleosomes expression in dermatomyositis (DM) and polymyositis (PM). To assess the aberrant expression of circulating nucleosomes in DM and polymyositis. 76 consecutive DM and PM patients from 1 September 2017 to 31 October 2018 were enrolled in this study, in addition to 20 healthy controls. The levels of circulating nucleosomes, with 16 kinds of detectable myo… Show more

“…Cell-free nucleosomes were measured using the Cell Death Detection ELISA Plus kit (Roche Diagnostics, Indianapolis, IN, US), which detects DNA and histones for specific mono- and oligonucleosomes detection, following the manufacturers’ instructions. Results are reported as a percentage of the positive control included in the kit as reported elsewhere ( 25 , 29 ).…”

“…Similarly, antiRo52 was found associated with RP-ILD and skin ulcers in anti-MDA5 patients (79). Serum markers were found able also to identify IIM subsets, as soluble interleukin-2R (sIL-2R) in discriminating DM from IMNM, particularly in case of active disease with ulcerative skin lesions (80), or neopterin in discriminating anti-MDA5 DM with severe ILD from other forms of IIMs (81); also, circulating nucleosomes, markers of cell apoptosis, were elevated in sera from IIMs patients compared to HCs, particularly in patients with anti-ARS, anti-MDA5 and anti-TIF1γ DM associated with skin involvement (82). Serum IL-17 levels were higher in ASS patients than in matched HCs, although apparently unrelated with the disease activity (83).…”

“…Quantification of cell-free nucleosomes was performed using the Cell Death Detection ELISA Plus kit (Roche Diagnostics, Indianapolis, IN, USA), which detects DNA and histones (antibodies from the mouse clones M-CA-33 and H11-4) for the specific detection of mono-and oligonucleosomes, following the manufacturers' instructions. Results are reported as a percentage of the positive control included in the kit as reported elsewhere [24].…”
