Abelson Interactor 1 (Abi1) and Its Interaction with Wiskott-Aldrich Syndrome Protein (Wasp) Are Critical for Proper Eye Formation in Xenopus Embryos

Abstract: Abl interactor 1 (Abi1) is a scaffold protein that plays a central role in the regulation of actin cytoskeleton dynamics as a constituent of several key protein complexes, and homozygous loss of this protein leads to embryonic lethality in mice. Because this scaffold protein has been shown in cultured cells to be a critical component of pathways controlling cell migration and actin regulation at cell-cell contacts, we were interested to investigate the in vivo role of Abi1 in morphogenesis during the developme… Show more

“…In addition, our results also clarified that this phenomenon is specific to NESH/Abi-3 among the three Abi proteins. Abi-1 is reported to be involved in the regulation of N-WASP/WASP [ 37 39 ], and other actin regulatory proteins such as Mena [ 6 ] and MIG10 [ 40 ]. It is believed that N-WASP/WASP is critical as an actin polymerization regulator for invadopodia formation.…”

“…Co-immunoprecipitation (co-IP) was performed according to (Singh et al, 2013). For each co-IP reaction, 10 μl reaction mix (66%) was mixed with 650 μl cold TNSG lysis buffer (20 mM Tris pH 8, 137 mM NaCl, 10% glycerol, 1% NP-40) containing a protease inhibitor cocktail tablet (Roche) and 2 μg/ml of anti-FLAG (Sigma), and incubated for one hour on ice.…”
