ABCD2 Alters Peroxisome Proliferator-Activated ReceptorαSignaling In Vitro, but Does Not Impair Responses to Fenofibrate Therapy in a Mouse Model of Diet-Induced Obesity

Abstract: Fenofibrate is a peroxisome proliferator-activated receptor (PPAR) a ligand that has been widely used as a lipid-lowering agent in the treatment of hypertriglyceridemia. ABCD2 (D2) is a peroxisomal long-chain acyl-CoA transporter that is highly induced by fenofibrate in the livers of mice. To determine whether D2 is a modifier of fibrate responses, wild-type and D2-deficient mice were treated with fenofibrate for 14 days. The absence of D2 altered expression of gene clusters associated with lipid metabolism, i… Show more

“…SNPs in genes in the common variant analysis with P < 1 × 10 ‐5 or q < 0.2 in the rare variant analysis were compared to gene expression results in wild‐type C57BL/6J mice fed vehicle control or fenofibrate for replication (REP1). Fifty‐seven genes overlapped between the two studies, and 10 were significant in REP1 ( q < 0.3; Mcm6 , Smad3 , Dcun1d4 , Hecw2 , Mipep , Ipo11 , R3hdm1 , Foxp1 , Stx8 , and Mapk10 ; Table ).…”

“…We investigated gene expression changes in wild‐type C57BL/6J mice administered fenofibrate compared with mice administered vehicle control, as described by Liu et al . to provide additional validation for common and rare variant associations with fenofibrate lipid response identified (REP1).…”

“…In spite of treatment of DIO rodents with fenofibrate has been reported to suppress weight gain and reduce visceral adiposity ameliorating metabolic disorder, the mechanisms by which fenofibrate exerts these effects have yet to be completely elucidated (Chaput, Saladin, Silvestre, & Edgar, ; Goto et al, ; Liu et al, ; Mancini et al, ; Rachid et al, ). The postulated mechanism involved in anti‐obesity effect of fenofibrate is a browning effect on subcutaneous fat depots (Rachid et al, ) via the increase in hepatic FGF21 (Goto et al, ).…”

“…ABCD2 plays a role in the degradation of long-chain saturated and omega 9monounsaturated fatty acids and in the synthesis of docosahexanoic acid (DHA) (Fourcade et al, 2009). The absence of ABCD2 altered expression of gene clusters associated with lipid metabolism, including PPARα signaling (Liu et al, 2014). Overexpression of ABCD2 alone prevented oxidative lesions to proteins in a mouse X-linked Adrenoleukodystrophy model (Fourcade et al, 2010) .…”
