A20 Inhibits LPS-Induced Inflammation by Regulating TRAF6 Polyubiquitination in Rainbow Trout

Abstract: The ubiquitin-editing enzyme A20 is known to inhibit the NF-κB transcription factor in the Toll-like receptor (TLR) pathways, thereby negatively regulating inflammation. However, its role in the TLR signaling pathway in fish is still largely unknown. Here, we identified a gene encoding A20 (OmA20) in rainbow trout, Oncorhynchus mykiss, and investigated its role in TLR response regulation. The deduced amino acid sequence of OmA20 contained a conserved N-terminal ovarian tumor (OTU) domain and seven C-terminal z… Show more

“…The supernatants containing secreted proteins were collected and filter-sterilized using a 0.22 µm filter. Samples were separated by SDS-PAGE using the PeptiGel TM I Peptide PAGE Analysis Kit (ELPIS-BIOTECH, Daejeon, Republic of Korea), and the immunoblotting assay was conducted as previously described [36]. The expressed AMPs were detected by incubation with a 6 × His-tag monoclonal antibody (Invitrogen) and a goat anti-mouse IgG (H + L) HRP-conjugated antibody (GenDEPOT, Katy, TX, USA).…”

“…The A20/AN1 zinc-finger protein's main role is to regulate the immune response. It also acts as a de-ubiquitinating enzyme, and is involved in controlling fundamental cellular activities [72][73][74]. Pentraxin, which functions as a soluble patternrecognition molecule, was also identified.…”

“…IRAK-M is another IRAK inhibitor, which belongs to the IRAK kinase family, but cannot induce NF-κB activation [ 82 ]. In addition to the inhibitors targeting IRAKs, proteins binding to TRAF6 [ 83 ], namely A20 [ 84 ] (also called TNF- α induced protein 3, TNFAIP3), NLR family member X1 (NLRX1) [ 85 , 86 ], and the cylindromatosis protein (CYLD), have also been shown to be negative regulators of TLR2 signaling [ 87 , 88 ]. Furthermore, A20 and NLRX1 can also block the activation of the IKK complex [ 89 ].…”
