A β‐mannan utilization locus in Bacteroides ovatus involves a GH36 α‐galactosidase active on galactomannans

Abstract: The Bacova_02091 gene in the β‐mannan utilization locus of Bacteroides ovatus encodes a family GH36 α‐galactosidase (BoGal36A), transcriptionally upregulated during growth on galactomannan. Characterization of recombinant BoGal36A reveals unique properties compared to other GH36 α‐galactosidases, which preferentially hydrolyse terminal α‐galactose in raffinose family oligosaccharides. BoGal36A prefers hydrolysing internal galactose substitutions from intact and depolymerized galactomannan. BoGal36A efficiently… Show more

“…As described above, a putative β-mannan PUL was previously identified in B. ovatus ATCC 8483 ( bacova_02087–02097 ) and shown to be transcriptionally up-regulated when galactomannan or glucomannan was included in the culture medium (10, 34). This PUL contains the genes bacova_02092 and bacova_02093 , which encode the putative GH26 β-mannanases Bo Man26A and Bo Man26B, respectively.…”

“…To further investigate the mechanism of β-mannan degradation by B. ovatus , we determined the cellular location of Bo Man26A, Bo Man26B, and the α-galactosidase Bo Gal36A, the latter of which carries a signal peptidase I site but no membrane-attachment motif (34). Custom antibodies to the three recombinant proteins were used to detect surface expression in formaldehyde-fixed non-permeabilized B. ovatus grown in minimal media containing LBG galactomannan as a sole carbon source.…”

“…A pair of xylan-targeting PULs from B. ovatus, PUL-XylS and PUL-XylL, was found to encode enzymes tailored for individual plant ␤-xylans that varied in their composition and branching (79). Recently, the detailed genetic, biochemical, and enzyme structural characterization of a galactomannanspecific PUL from B. ovatus revealed the interplay of two mannan-specific SGBPs, two GH26 endo-␤-mannanases, and a GH36 exo-␣-galactosidase in the deconstruction of this plant cell wall polysaccharide (110,111). Among environmental bacteria, a complex chitin utilization locus from Flavobacterium johnsoniae has been extensively functionally characterized (112).…”
