A zinc complex that suppresses the expression of a reactive sulfur species-producing enzyme, cystathionine γ-lyase, in cultured vascular endothelial cells

Abstract: Persulfide species present in mammalian cells play important toxicological roles against oxidative stress and methylmercury. Cystathionine γ-lyase (CSE) and cystathionine β-synthase (CBS) are enzymes that produce persulfide species using cysteine as a substrate in the presence of vitamin B 6. However, little is known about the regulatory mechanisms underlying the expression of these enzymes. In the present study, we searched for molecular probes from a library of 27 zinc complexes to analyze the mechanisms in … Show more

“…As the creation of such nanomaterials has become widespread, the toxicity and distribution of the molecules have also been studied (Perrault et al, 2009;Chou and Chan, 2012;Jiang et al, 2015;Foroozandeh and Aziz, 2018). We also reported the cytotoxicity and function of organic-inorganic hybrid molecules -metal complexes and organometallic compounds-as tools for analyzing biological systems of vascular endothelial cells at different cell densities (Fujie et al, 2016(Fujie et al, , 2019(Fujie et al, , 2020Hara et al, 2017Hara et al, , 2018aHara et al, , 2019Hara et al, , 2020Takahashi et al, 2018). However, because these hybrid molecules are generally electrophilic, the response of these cells against metal compounds that lack electrophilicity is not well understood.…”

“…Smad2/3 and β-actin proteins were separated by SDS-PAGE, and western blotting was performed as described previously (Takahashi et al, 2018). Immunoreactive bands were visualized using Chemi-Lumi One Super western blot detection reagent and scanned using an Amersham Imager 600 (GE Healthcare).…”
