A Vesicular Transport Pathway Shuttles Cargo from Mitochondria to Lysosomes

Abstract: Mitochondrial respiration relies on electron transport, an essential yet dangerous process in that it leads to the generation of reactive oxygen species (ROS). ROS can be neutralized within the mitochondria through enzymatic activity, yet the mechanism for steady-state removal of oxidized mitochondrial protein complexes and lipids is not well understood. We have previously characterized vesicular profiles budding from the mitochondria that carry selected cargo. At least one population of these mitochondria-der… Show more

“…56 Overall, it is clear that multiple specific molecular pathways exist for targeting damaged or unneeded mitochondria for selective clearance by autophagy. A given stimulus, such as CCCP, can trigger more than one pathway to include the canonical Pink1/Parkin pathway, a vesicular lysosomal-targeting pathway, 57 and the NDPK-Ddependent CL externalization pathway described here. Based on the comparison of the data on the effects of NDPK-D on CL translocation with those illustrating the role of NDPK-D in Parkin translocation, we suggest that these processes may be related to each other.…”

“…In yeast, V-ATPase mutations have an impact on petite phenotypes (44,45), and V-ATPase function has also been linked to cellular aging and rejuvenation by controlling mitochondrial function (46). In human cells, vesicular transport operates between lysosomes and mitochondria (47). Indeed, there is growing evidence supporting roles for lysosomes in signaling and energy metabolism beyond their established roles in degradation and recycling (48).…”

“…6 In addition to proteolytic and proteosomal degradation, recent evidence points to a lysosomal pathway in which vesicles bud from mitochondrial tubules, sequester selected mitochondrial cargos, and then deliver those mitochondrial components to the lysosome for degradation. 7 This pathway is active under steady-state conditions and is further stimulated by oxidative stress. These mitochondrially derived vesicles may represent a mechanism for selective removal of oxidized mitochondrial proteins while leaving the whole organelle intact.…”
