A validated method for quantitation of psilocin in plasma by LC–MS/MS and study of stability

Abstract: A liquid chromatography-electrospray ionization/tandem mass spectrometry method for the quantitation of psilocin in plasma is presented. Sample workup was performed with mixed-mode solid-phase extraction using ascorbic acid and nitrogen for drying to protect the unstable analyte. Calibration curves were linear from 2 to 100 ng/mL, and no selectivity problems occurred. The limit of detection was 0.1 ng/mL, and the limit of quantitation was 0.34 ng/mL. Recovery was >86% and matrix effects were <110%. Both were r… Show more

“…Nonetheless, it has to be said that both scientific reports and websites which commercialize "magic truffles" claimed that psilocybin and psilocin are light and temperature-sensitive and that degradation of the two alkaloids, particularly psilocin, may occur if mushrooms (and consequently sclerotia) are not stored at low temperature or shipped in vacuum. [8,15,19,20] Usually bluing of mushrooms corresponds to alkaloids oxidation and consequent degradation. [8,19] This was what probably occurred with sclerotia seized the Italian anti-adulteration and safety bureau (Carabinieri per la tutela della salute-NAS).…”

“…cathinones, 26,27 piperazines, 27,28 and synthetic cannabinoids. 29 Martin et al 30 could overcome the psilocin instability in plasma by freezing. In contrast, psilocin glucuronide, the main psilocin metabolite, was stable in serum and urine and should thus be included in the approach for monitoring psilocin intake.…”

“…18 Psilocin and bufotenine glucuronide should be included as psilocin-O-glucuronide showed a higher stability in biosamples amongst others and as it can be distinguished from bufotenin-O-glucuronide via their MS 2 patterns. 18,29,30 A strategy should be developed and validated offering an easier of workflow and less turnaround time. Applicability should be tested by analyzing human urine samples obtained from suspected mushroom intoxication cases.…”
