Over the years, Newcastle disease (ND) has defied all available control measures. The disease has remained at the forefront of infectious diseases afflicting poultry production after avian influenza. Despite the continuous global use of million doses of ND vaccine annually, the causative pathogen, avian paramyxovirus type 1 also known as Newcastle disease virus (NDV) has continued to evolve causing, even more, a threat not only to the unvaccinated but the vaccinated flocks inclusive. The disease has been well studied in the developed countries where the virus is found in circulation. However, limited information exists on the epizootiology and circulating genotypes of the virus in developing countries where the majority of the flocks are raised on the extensive management system. Identification of virulent NDV in apparently healthy free-range ducks in this system calls for concern and pragmatic approach to investigate factor(s) that favour the virus inhabiting the ducks without clinical manifestation of the disease. Recently, novel genotypes (XIV, XVII, and XVIII) with peculiarity to West and Central African countries have been discovered and due to lack or poor surveillance system possibility of hitherto unreported genotypes are likely. This review elucidates and discusses available literature on the diversity of the circulating NDV genotypes across the West Africa countries and the epizootiology (molecular) of the disease in Nigeria with the view of identifying gaps in knowledge that can assist in the development of effective vaccines and control strategies to combat the peril of the disease.

Newcastle disease is a highly contagious disease responsible for major outbreaks and considerable economic losses in the poultry industry in China. There is still little information available regarding gene characterization of the NDV, especially in ducks and pigeons. Therefore, the aim of this study was to investigate NDV isolated from ducks and pigeons in Hubei, China. In this study, three NDVs from ducks and pigeons were isolated between 2013 and 2015.The fusion protein (F) gene of the NDV isolates was sequenced and phylogenetically analyzed. The clinical signs and gross histopathological lesions were examined. Phylogenetic analysis of these strains indicated that all the sequences are classified as genotype II. The isolates shared a 112 G-R-Q-G-R-L 117motif at the F protein cleavage site, indicating that these three isolates strains are lentogenic. Necropsy and histopathology showed the typical pathological changes. It was concluded that commercial ducks and pigeons in Hubei province carry lentogenic NDV strains with regular genetic divergence, indicating that these species may act as the main reservoirs of NDV in poultry. Therefore, strategies and surveillance should be undertaken to reduce the risk of ND outbreaks.

Newcastle disease (ND) is one of the highly contagious disease responsible for devastating outbreaks and considerable economic losses to poultry industry in China. However, no information is available about NDV in Tibet region; the aim of this study was to determine the genetic characterization and evolutionary analysis of NDV in Tibetan chickens. Four NDVs were isolated from an outbreak in Tibetan chickens. The pathogenicity of the isolates was determined by mean death time (MDT), intracerebral pathogenicity index (ICPI) and amino acid sequencing (112 to 117) of F protein. While the genetic characterization of F gene was determined by sequencing the isolated NDVs and phylogenetic relationship was established with the help of reference strains. Pathogenicity experiments revealed that XZ-F10 and XZ-F20 strains were lentogenic pathotype, while the remaining XZ-F2-1 and XZ-F4-6 strains were velogenic pathotype. The deduced amino acid sequences of the cleavage site of the F protein confirmed our results. Phylogenic analysis of these strains indicated that both XZ-F2-1 and XZ-F4-6 belong to genotype VII. However, XZ-F10 and XZ-F20 strains were assigned to genotypes II. The present study highlights the need for continuous surveillance of NDV in Tibetan chickens; moreover, this study will provide a reference for the local government to make the strategies and control emerging NDV in Tibet.
