A unique human ovarian carcinoma cell line expressing CD34 in association with selection for multidrug resistance

Abstract: A new cell line of nonhematopoietic and nonvascular endothelial origin that expresses CD34 in association with selection for MDR was cloned. A study of MDR mechanisms in this cell line revealed that reduced type II alpha topoisomerase levels were likely responsible for the MDR observed. A study of the causal relation between the selection of drug resistance and the expression of CD34 may provide insight into why CD34 correlates with poor clinical response in patients with acute myeloid leukemia.

“…The characteristics of the human ovarian cancer cell line A2780/wt (parental), the MDR subline A2780/Dx5 (P-gp-positive, MRP-negative), the human breast cancer cell line MCF-7/wt (parental) and the MDR subline MCF-7/adr (P-gp-positive, MRPnegative) have been reported previously (Fairchild et al, 1987;Minderman et al, 1996). Parental and resistant A2780 and MCF-7 cells were grown as monolayer in RPMI-1640 medium supplemented with 10% heat-inactivated fetal calf serum (FCS), and 2 mM L-glutamine.…”

“…The human ovarian carcinoma cell lines A2780 and A2780-Dx5b [15] represent Pgp-mediated MDR with the A2780-Dx5b cell line displaying a 35-fold resistance to doxorubicin compared to its parental cell line. The human ®bro-sarcoma cell lines HT1080 vs HT1080/Dr4 [19,20] represent MRP-mediated MDR with a 180-fold resistance to doxorubicin of the HT 1080/Dr4 cell line compared to the HT 1080 cell line.…”

“…The adherent Pgp expressing ovarian carcinoma cell line A2780/dox5 16,17 and the leukemia cell line HL60/ADR [18][19][20] displaying the MRP1 phenotype and their parent cell lines A2780/WT and HL60/WT were cultured in Iscove's medium (Life Technologies, Paisley, UK) supplemented with 10% heatinactivated fetal calf serum (FCS; Hyclone, Logan, UT, USA), 2 mM glutamine and 100 IU/ml penicillin, at 37°C in a humidified atmosphere with 5% CO 2 . The cells were used in an exponential growth phase and all cultures were routinely checked for mycoplasma contamination.…”
