A unified model of the hierarchical and stochastic theories of gastric cancer

Abstract: Gastric cancer (GC) is a life-threatening disease worldwide. Despite remarkable advances in treatments for GC, it is still fatal to many patients due to cancer progression, recurrence and metastasis. Regarding the development of novel therapeutic techniques, many studies have focused on the biological mechanisms that initiate tumours and cause treatment resistance. Tumours have traditionally been considered to result from somatic mutations, either via clonal evolution or through a stochastic model. However, em… Show more

“…The cancer problem has been considered from many different modeling perspectives ranging from the standard mechanistic models [21][22][23][24][25], to applications of population dynamics [26][27][28][29], evolutionary game theory [30][31][32], and models of swarm dynamics [33,34]. However, these methods generally only consider one or two levels of organization in describing the dynamics of the systems they consider, and almost exclusively view the lowest level as giving rise to the higher levels.…”

“…In addition to transcription factors (TFs) (Finley et al, 2018;Niwa, 2018) and cell surface markers (Sahlberg et al, 2014), miRNAs are increasingly recognized as potential stem cell markers (Lou et al, 2018;Song et al, 2017). The miRNA expression, as with other genes, is under the control of TFs and their genes may be transcribed by RNA polymerase II or III.…”

“…Song et al . recently proposed a unified model for GC, in which primitive GCSCs continually optimize their genome, improve the pattern of gene expression, and generate various subclones to adapt to the changing tumor microenvironment.…”
