A Tyrosine-Based Signal Targets H/K-ATPase to a Regulated Compartment and Is Required for the Cessation of Gastric Acid Secretion

Abstract: Gastric acid secretion is mediated by the H/K-ATPase of parietal cells. Activation of acid secretion involves insertion of H/K-ATPase into the parietal cell plasmalemma, while its cessation is associated with reinternalization of the H/K-ATPase into an intracellular storage compartment. The cytoplasmic tail of the H/K-ATPase beta subunit includes a four residue sequence homologous to tyrosine-based endocytosis signals. We generated transgenic mice expressing H/K-ATPase beta subunit in which this motif's tyrosi… Show more

“…The presence of these and other potential endocytosis motifs in coronin se is particularly intriguing given recent findings that alteration of a tyrosine-based endocytosis motif within the ␤-subunit of the H ϩ ,K ϩ -ATPase leads to constitutive acid secretion in transgenic mice (37). Finally, since PKC activation has been associated mainly with the inhibition of HCl secretion (22,38), increases in coronin se phosphorylation might also serve to increase the endocytotic retrieval of the ion-transporting H ϩ ,K ϩ -ATPase or some other secretion-related ion channel or transporter from the apical membrane.…”

“…Briefly, HA-Y543-expressing cells grown to confluency on Transwell filters were incubated at 37°C for 40 min in Met-Cys-free medium and labeled for 30 min with 35 S-Met and Cys (ICN Radiochemicals, Irving, CA). Cells were chased with complete medium for 2 h at 37°C.…”

“…The ␣-subunit contains ATP and cation binding sites and carries out the catalytic and transport functions of the enzyme (1), and it also contains sequences responsible for apical membrane localization (4). The heavily glycosylated ␤-subunit is required for endocytic retrieval of the H,K-ATPase from the canilicular membranes as the cell passes from the stimulated to the resting state (5) and may also contribute to proper folding and membrane localization of the enzyme (1).…”
