A Two-Photon Fluorescent Probe for Lysosomal Thiols in Live Cells and Tissues

Abstract: Lysosome-specific fluorescent probes are exclusive to elucidate the functions of lysosomal thiols. Moreover, two-photon microscopy offers advantages of less phototoxicity, better three dimensional spatial localization, deeper penetration depth and lower self-absorption. However, such fluorescent probes for thiols are still rare. In this work, an efficient two-photon fluorophore 1,8-naphthalimide-based probe conjugating a 2,4-dinitrobenzenesulfonyl chloride and morpholine was designed and synthesized, which exh… Show more

“…However, since NIR‐excited NPs are effectively applied in vivo for thermo‐chemotherapy and PDT, what advantages do TPE‐NIR cancer treatments provide? TPE‐NIR has an unparalleled 3D spatiotemporal resolution at the laser focal volume (1 µm 3 ), 2 cm deep into tissues to eradicate cancer cells . Larger tumors can be treated accurately via a laser screening of the diseased organs and tissues in several minutes.…”

“…Herein, photoCORMs compounds 1 – 2 (Scheme ) combine a [Mn I (CO) 3 ] CO‐rich fragment with a potentially emissive dipicolylamine‐1,8‐naphthalimide ligand, while 2 also integrates a biotin tether. The turn‐on fluorescent chromophore 1,8‐naphthalimide has a large 2PA cross‐section and can be excited between λ exc =600–950 nm . Biotin is required to sustain the natural growth of eukaryotic cells .…”

“…The use of the reactivity‐based chemical probes is particularly advantageous, because they provide a highly sensitive means to detect biothiols with excellent specificity. Moreover, thiol‐responsive fluorescent probes can be used for live ‐ cell imaging application, which enables dynamic monitoring of the thiol distribution in the intracellular environment …”
