A transplantable insulinoma in the rat.

Abstract: A transplantable insulinoma was developed in inbred albino rats of the NEDH strain. The original tumor, 1 cm in diameter, was removed from the pancreas of a male parabiont 566 days following 1000 rads (10 J/kg) of total body x-irradiation. The time required for implanted fragments to grow to 0.5-1.5 cm in diameter decreased from 5-8 months in the first generation to 2-5 months in the seventh generation. Successful transplantation in male animals followed for 4 or more months after transplantation was significa… Show more

“…The tumour, if shown capable of the exocytosis of insulin, would provide secretory material in greater quantity than is obtainable by isolating islets; and of far higher insulin content and degree of differentiation than that from previously studied islet cell tumours [2,9,10]. The characteristics of insulin secretion by pancreatic B cells have been described in detail [for reviews see 3, 8].…”

“…RINm5F cells of the insulin-producing cell line, derived from a rat islet cell tumor [22,23], were grown in RPMI-1640 medium containing 10% (v/v) heat-inactivated fetal calf serum, 50 IU/ml penicillin, 0.25 /xg/ml fungizone and 50 ixg/ml streptomycin at 37°C in an atmosphere of humified air/CO 2 (19:1) according to Praz et al [24] with the following modifications: The cells were seeded at a density of (2-4)" 104 cells/ml in 20 ml of medium (75 cm 2 culture flasks). The medium was replaced four times per week (one passage).…”
