A tissue culture technique for rapid clonal propagation and storage under minimal growth conditions of Musa (Banana and plantain)

Abstract: A tissue culture technique for rapid clonal propagation and storage under minimal growth conditions is presented in this paper. Shoot-tip cultures of Musa cultivars (both banana and plantain) are induced by culturing small excised shoot apices on modified MS semisolid medium supplemented with various concentrations and combinations of auxins and cytokinins. The effects of cytokinin concentration in the medium as well as the genotypic configuration of the cultivars on the rate of shoot-bud proliferation have be… Show more

“…Micropropagation techniques were developed during the past two decades and are now well established (Banerjee and De Langhe 1985;Cronauer and Krikorian 1984;Israeli et al 1995;Vuylsteke 1989). Tissue culture labs supply the regular replanting needs of the large export banana plantations (Smith and Drew 1990a).…”

“…Shoot-tip cultures maintained at lower temperatures (15-18°C) have been used for the slow-growth storage of Musa germplasm (Banerjee and De Langhe 1985;Van den houwe et al 1995). A simple technique was recently developed for cryopreservation of meristem cultures, which involves preculture on high-sucrose medium followed by rapid freezing (Panis et al 1996).…”

“…uniformity of plants, freedom from viral diseases, plants give an early yield, easy for bulk transportation, faster turnover/mass production of plants all the year round. However, during field establishment, the tissue culture plants require a higher level of care than conventional planting material (Banarjee and De Langhe, 1985).…”
