Isoelectric focusing (IEF) of native sera from immature or adult rats and of purified or partially purified rat serum thyroid hormone-binding proteins, demonstrates that rat TBG is a microheterogeneous protein. Autoradiography and radioactivity scans of the IEF plates show that it consists of at least four main isoforms, with bands at pH 4.35, 4.45, 4.5 and 4.55. This pattern is remarkably similar to that reported for human TBG. “…Rodents contain a gene that can encode the TBG protein, but it is expressed at very low levels in adult animals (Vranckx et al, 1990;Rouaze-Romet et al, 1992;Tani et al, 1994). Developmental studies show TBG protein levels increase during the early postnatal period in rodents, but then decline to very low levels by weaning and remain low through the remainder of the animal's life span (Savu et al, 1987(Savu et al, , 1991Vranckx et al, 1990). As in humans, T3 and T4 have higher affinity for TBG than TTR or albumin in rodents.…”

The highly conserved nature of the thyroid gland and the thyroid system among mammalian species suggests it is critical to species survival. Studies show the thyroid system plays a critical role in the development of several organ systems, including the reproductive tract. Despite its highly conserved nature, the thyroid system can have widely different effects on reproduction and reproductive tract development in different species. The present review focuses on assessing the role of thyroid hormones in human reproduction and reproductive tract development and comparing it to the role of thyroid hormones in laboratory animal reproduction and reproductive tract development. The review also assesses the effects of thyroid dysfunction on reproductive tract development and function in humans and laboratory animals. Consideration of such information is important in designing, conducting, and interpreting studies to assess the potential effects of thyroid toxicants on reproduction and development.

Approved for public release; distribution is unlimited. ABSTRACT (Maximum 200 words)Perchlorate interferes with iodide accumulation in the thyroid resulting in reduced thyroidal hormone synthesis. The objective of this project was to conduct a series of experiments to investigate the role of perchlorate on the uptake of iodide in the thyroid and its effects on the synthesis of thyroid hormones. A time course study with perchlorate (single iv dose) was performed to investigate its kinetic behavior in the adult male rat. Rats were injected with 0, 0.01, 0.1 1 and 3 mg perchlorate/kg via tail vein, challenged with 1251 with carrier at 2 h post dosing, and euthanized at predetermined time points. Three groups (n=6 per group) of 1-, 5-, and 14-day drinking water studies were performed at different levels of perchlorate (0, 1, 3 and 10 mg/kg). At the end of exposure, rats were challenged once by tail vein injection with 1251 with carrier. Dose-related inhibition of thyroid iodide uptake after a single iv dose of perchlorate, and both dose-and timedependent changes in inhibition of thyroid iodide uptake were observed in drinking water studies. In both the single dose and drinking water studies, TSH and free T4 levels were elevated with T4 levels decreased. Serum T3 levels in drinking water studies remained unchanged in all dosing groups.
