A third booster dose may be necessary to mitigate neutralizing antibody fading after inoculation with two doses of an inactivated SARS‐CoV‐2 vaccine

Abstract: this study has many limitations, such as the sample size, age group, sex composition, and physical characteristics. Continuously observing the persistence of the protection provided by vaccines in real cases and the effectiveness of a booster dose (a third dose), conducting long-term clinical trials, and obtaining post-clinical data are essential tasks. To some extent, this study showed that serum neutralizing antibody levels decreased after the second dose of inactivated vaccines. Importantly, the results sugg… Show more

“…Inactivated vaccines provoke an immune response by humoral immunity, therefore, they require multiple doses [ 15 ]. A third booster is encouraged in China to induce a stronger protection [ 16 ]. Commonly proposed mechanisms of inflammation may be induced by the adjuvants and have been classified as Shoenfeld syndrome typically occurring in patients with an autoimmune disorder, often accompanied by arthralgia and fatigue [ 17 ].…”

“…Different types of COVID-19 vaccines including mRNA, viral vector, inactivated, and protein-based vaccines are used as boosters ( 33 , 36 , 38 41 ). In general, a booster dose can substantially increase NAb levels or their surrogate markers (e.g., anti-spike IgG) against SARS-CoV-2.…”

“…Recently, the need for a third dose of the COVID-19 vaccine has been discussed due to the threat of the B.1.617.2 (delta) variant and the need for the protection of immunocompromised individuals [13]. A three-dose regimen could reduce the risk of COVID-19 infection and reverse the waning of neutralizing antibodies after two doses of the vaccine [14][15][16]. However, the best timing of a third dose or boosting with heterologous vaccines is controversial [13,17,18].…”
