A termination-independent role of Rat1 in cotranscriptional splicing

Abstract: The yeast termination factor Rat1, and its human homolog Xrn2, have been implicated in multiple nuclear processes. Here we report a novel role of Rat1 in mRNA splicing. Rat1 mutants display increased levels of unspliced transcripts. Accumulation of unspliced transcripts was not due to a failure to degrade unspliced mRNA, disruption of termination or an increased elongation rate in Rat1 mutants. ChIP-Seq analysis revealed Rat1 crosslinking to the introns of a subset of yeast genes. Mass spectrometry and coimmun… Show more

“…Exoribonuclease Rat1 trims cleaved pre-rRNA fragments 38 and degrades telomeric repeat-containing RNAs (TERRA) 39 . It also promotes transcription termination of RNAPI 40 and RNAPII 41,42 , contributes to the initiation-elongation transition 43 , mRNA splicing 44 , and the decay of uncapped or splicing-defective mRNAs 44,45 . Strong genetic interactions between mutants in Rat1 and myriad kinetochore proteins 46 suggested an involvement of this RNase in CEN transcript turnover.…”
