The endocannabinoid system (eCB) began to be studied from the identification of the molecular structures present in the cannabis sativa plant. The ECS is constituted of cannabinoid receptors, endogenous ligands and all the associated enzymatic apparatus responsible for maintaining homeostasis. Several physiological effects of cannabinoids are exerted through interaction with various receptors such as CB1 and CB2 receptors, vanilloid receptors, and the recently discovered [GPCRs (GPR55, GPR3, GPR6, GPR12 and GPR19). Endogenous ligands such anandamide and 2-arachidonoylglycerol might modulate these receptors. eCB has proved to play a critical role in some human diseases and has been extensively studied due to its wide therapeutic potential and because it is a promising target for the development of new drugs. Phytocannabinoids and synthetic cannabinoids have shown varied affinities to eCB, which are relevant to the treatment of various diseases. They may also have potential as lead compounds in the development of cannabinoid-based pharmaceuticals for a variety of diseases. Furthermore, Integrative and Complementary Health Practices (ICHP) appear to influence the endocannabinoid system through modulation. This review will show a description of ECS components and discuss how phytocannabinoids, other exogenous compounds, and PICS may operate the eCB balance.

Badanie z 2018 roku przeprowadzone na 20 pacjentach, w którym wykorzystano trzy odmiany marihuany podawane w formie inhalacji -Bedrocan (22.4 mg THC, <1 mg CBD); Bediol (13.4 mg THC, 17.8 mg CBD), Bedrolite (18.4 mg CBD, <1 mg THC) -wykazało redukcję bólu.…”

Introduction and purpose: Fibromyalgia is a disease with many symptoms that are difficult to treat. The most troublesome element is muscle, bone or joint pain. Importantly, the diagnosis of this disease is based on the physical examination and the subjective examination, which shows that there are no laboratory tests to confirm its presence. Public awareness of fibromyalgia is negligible, while more research is needed on the treatment of this disease and the possible alternatives associated with it. The aim of this study was to analyze the available literature on the basis of the PubMed scientific database, considering only scientific articles that were published within the last 5 years. The following keywords were used: fibromyalgia and physiotherapy, fibromyalgia and cannabis, fibromyalgia and cytokines. Description of the state of knowledge: Treatments related to cannabinoids and physiotherapy are still developing fields of knowledge. More than 40 papers related to the topic of the study were used for the review. The need for further research is apparent, especially randomized studies in which allocation to an experimental or control group is completely random. The question of investigating the long-term effectiveness of a given treatment is also not negligible, as many studies do not have a so-called follow-up. Conclusion: Cannabinoids are effective in the treatment of complaints associated with fibromyalgia. Exercise, laser therapy and acupuncture reduce pain and improve quality of life in patients with fibromyalgia. More research on this disease is needed, especially with larger patient groups.

A Review on the Bioactivity of Cannabinoids on Zebrafish Models: Emphasis on Neurodevelopment

For centuries, the cannabis plant has been used as a source of food, fiber, and medicine. Recently, scientific interest in cannabis has increased considerably, as its bioactive compounds have shown promising potential in the treatment of numerous musculoskeletal and neurological diseases in humans. However, the mechanisms that underlie its possible effects on neurodevelopment and nervous-system functioning remain poorly understood and need to be further investigated. Although the bulk of research on cannabis and cannabinoids is based on in vitro or rodent models, the zebrafish has now emerged as a powerful in vivo model for drug-screening studies and translational research. We here review the available literature on the use of cannabis/cannabinoids in zebrafish, and particularly in zebrafish models of neurological disorders. A critical analysis suggests that zebrafish could serve as an experimental tool for testing the bioactivity of cannabinoids, and they could thus provide important insights into the safety and efficacy of different cannabis-extract-based products. The review showed that zebrafish exhibit similar behaviors to rodents following cannabinoid exposure. The authors stress the importance of analyzing the full spectrum of naturally occurring cannabinoids, rather than just the main ones, THC and CBD, and they offer some pointers on performing behavioral analysis in zebrafish.
