A Systematic Approach for Liposome and Lipodisk Preclinical Formulation Development by Microfluidic Technology

Abstract: Lipid nanoparticles have transformed the drug delivery field enhancing the therapeutic drug performance of small molecules and biologics with several approved drug products. However, in industry, these more complex drug delivery systems such as liposomes require more material and time to develop. Here, we report a liposome and lipodisk decision tree with model compounds of diverse physicochemical properties to understand how to resourcefully optimize encapsulation efficiency (EE) for these lipid-based drug del… Show more

“…The conventional liposomes were prepared through a microfluidics method with the NxGen Blaze (Precision NanoSystems, Vancouver, BC, Canada) system, as previously described in [ 23 ]. First, the DSPC, DSPE-PEG2000, and cholesterol (61.56%:18.8%:19.56% ( w/w )) were dissolved, at a total lipid concentration of 30 mg/mL, in ethanol.…”

“…6V). 51,53 This technology enables the production of small liposome nanoformulations with precise control of their mixing, size, distributions, and fluid flow rate. The main advantages of this technique are its good particle size control, the ability to produce almost all types of vesicles such as SMVs, SUVs, GUVs, and LUVs, and the possibility of upgrading to novel methods for liposome preparation.…”

“…It has the advantage of basically no preprocessing, no advance normalization, and handling missing values. 77 However, it is very prone to overfitting, resulting in poor generalization, and it is difficult to learn for some of the more complex relations. Trinh et al used in vitro characteristics as input parameters (for example, cell line, source, type, and so on).…”
